DESCRIPTION:Cellular and molecular mechanisms driving aberrant development in a novel model of Venous Malformation \n\n\n\n Patients with venous malformation (VM) develop vascular lesions consisting of massively enlarged and dysmorphic veins. These lesions cause significant disfigurement\, chronic pain\, and complications such as bleeding and coagulopathy. VMs are associated with somatic\, gain-of-function mutations in the endothelial cell receptor kinase TIE2. The cellular and molecular mechanisms by which TIE2 mutations drives formation of VM are still not fully understood. DESCRIPTION:Lymphatic endothelial cell cycle control enables lymphatic valve formation \n\n\n\nThe lymphatic vasculature relies on intraluminal valves to maintain unidirectional lymph flow and tissue fluid homeostasis. Disruption to their formation during embryonic development leads to congenital lymphedema\, or aberrant pooling of lymph in the tissues leading to tissue swelling\, fibrosis\, and localized immune dysfunction. Despite this\, our understanding of the mechanisms that regulate lymphatic valve formation is not complete. DESCRIPTION:Nanoscale Materials to Target Diseases in the Vasculature \n\n\n\nCardiovascular disease is the leading cause of mortality globally\, with inflammation-inducing atherosclerotic plaque being the main contributing factor. The CANTOS trial showed that decreasing inflammation reduced cardiovascular events without lowering lipids\, but systemic anti-inflammatory administration was limited by off-target effects. Thus\, there is a great need for a new therapeutic that actively prevents progression of atherosclerotic plaques by modulating inflammatory pathways. Nanotechnology offers a noninvasive approach for the management of cardiovascular disease via targeted delivery of therapeutic agents that are otherwise limited by pharmacokinetics or toxicity from systemic administration. We have engineered a peptide amphiphile (PA) specifically targeted to sites of atherosclerosis by covalently incorporating an apolipoprotein A1 (ApoA1)-mimetic peptide. We showed that these PAs self-assemble into supramolecular nanofibers that localize to sites of atherosclerosis when intravenously administered to low-density lipoprotein receptor knock out (LDLR-/-) mice.
