DESCRIPTION:Cavitation-facilitated AAV Gene Therapy for Cardiovascular and Muscular Disorders (Maya) \n\n\n\nDruggable Pathways Involving Platelet Signaling for Slowing Aortic Stenosis Progression (Ruie-Jen) \n\n\n\nIn the first half of the series\, we will review progress to date on augmenting AAV gene therapy by ultrasound cavitation of microbubble contrast agents. We will discuss work performed to optimize acoustic environment\, to understand mechanism\, and to translate the technology from cardiac to skeletal muscle. In the second half of the series\, we will review current progress on understanding the role of platelets in promoting aortic stenosis (AS) progression through myofibroblastic and osteogenic transformation\, and EndoMT of valve endothelial cells. DESCRIPTION:Contributions of hypertension to cognitive decline and Alzheimer’s disease  \n\n\n\nCardiovascular risk factors are now recognized as important contributors not only to vascular dementia but also to Alzheimer’s disease. Indeed\, there is a strong association between mid-life hypertension and late-life dementia. My lab seeks to understand the cellular and molecular mechanisms underlying the detrimental effects of hypertension on cognition and brain health by using preclinical models to answer these questions. We have previously identified novel role of neuroimmune activation in the dura contributing to the impairment of cerebral blood flow regulation and cognitive function in hypertension. DESCRIPTION:Cellular and molecular mechanisms driving aberrant development in a novel model of Venous Malformation \n\n\n\n Patients with venous malformation (VM) develop vascular lesions consisting of massively enlarged and dysmorphic veins. These lesions cause significant disfigurement\, chronic pain\, and complications such as bleeding and coagulopathy. VMs are associated with somatic\, gain-of-function mutations in the endothelial cell receptor kinase TIE2. The cellular and molecular mechanisms by which TIE2 mutations drives formation of VM are still not fully understood. DESCRIPTION:Lymphatic endothelial cell cycle control enables lymphatic valve formation \n\n\n\nThe lymphatic vasculature relies on intraluminal valves to maintain unidirectional lymph flow and tissue fluid homeostasis. Disruption to their formation during embryonic development leads to congenital lymphedema\, or aberrant pooling of lymph in the tissues leading to tissue swelling\, fibrosis\, and localized immune dysfunction. Despite this\, our understanding of the mechanisms that regulate lymphatic valve formation is not complete.
